Each game of Heroes of Newerth pits two teams of 1 to 5 players, called the Legion and Hellbourne, against each other. Each player selects a single hero to play from a pool of sixty, via one of several selection rotations. During the match, heroes will gain experience and gold by destroying enemy NPC units, players or structures, and customize their character with skills and items as the game progresses. Gameplay consists of heroes attacking/defending NPCs in a "lane" - a pre-defined pathway from one base to the other that the AI-controlled NPCs follow - to gain experience and gold advantage over the other team, while concurrently attempting to kill opposing heroes and avoid being killed themselves, to respawn a short time later in their own base. A game ends either when one of the teams' main structure is destroyed - either the World Tree for the Legion, or the Sacrifical Pit of the Hellbourne. A team may also concede at any point beyond fifteen minutes into the match.

Currently there are a total of sixty heroes consisting of thirty for each faction. Heroes are additionally organized by their main attribute - either strength, intelligence, or agility. A main attribute, along with the set of abilities that is provided for a hero, loosely assigns the role that they will fill in the game - such as a "tank" who is able to absorb a lot of damage, "carry" who scales very well the longer the game moves on and deals massive damage, "initiator" who starts "team fights" or "clashes" and last "support" these heroes have a very wide range of abilities that range from healing and buffing to damage and disables. Each hero in the game has four unique abilities that can be learned as levels are gained from the death of opposing heroes or creeps. Some abilities are active and must be manually executed, while some are passive and are often left to chance.

Hero selection is dependent on the type of game being played. The main game types are Normal, in which players can select from the entire pool of heroes from their faction; Single Draft, in which a player is assigned three heroes, one hero of each attribute type, to select from; Random Draft, in which players alternately pick from the same random pool of heroes from both factions; Banning Draft, which is similar to Random Draft, with each team captain (this is assigned by the player slots, blue and pink) first disqualifying two heroes from being selected by anyone; and Deathmatch, which is currently disabled. An extra option called "All Heroes" is available, which modifies all the game modes to allow the heroes to come from the entire pool of both factions, rather than just the side the player is on.http://www.alienwarearena.com/event/hon-beta-giveaway
